Well that's because you are missing a "c". This works:
\documentclass{amsart}
\makeatletter
\renewcommand*\@acciii{\mathbin{\,=\,}}
\renewcommand*\={\@acciii}
\makeatother
\begin{document}
\[ A\= B \]
\end{document}
